Research Interests

Parturition

Preterm labor is a major problem in Obstetrics and its incidence has not been reduced in the past 20years. Studies in our laboratory are intended to define the molecular mechanisms occurring in labor at term and preterm and will hopefully lead to better methods to regulate preterm (and term labor). Prostaglandins are intimately involved in parturition and the regulation of prostaglandin synthesis , metabolism and action is the focus of our studies.

Regulation of PGHS-2 and PGDH expression in Human Pregnancy

On-going studies are designed to define the mechanism of glucocorticoid regulation of PGHS-2 expression, (the major enzyme involved in PG formation in pregnancy) and PGDH the major enzyme involved in the regulation of PG degradation. The role of glucocorticoid and progesterone receptors together with their co-activators and co-repressors in the regulation of PGHS-2 and PGDH in the human fetal membranes is being examined.

Regulation of Prostaglandin Production and Action in Pregnancy and Parturition

We have recently proposed a new model for the regulation of parturition in the sheep, whereby fetal glucocorticoids increase expression of PGHS-2 in the fetal placenta resulting in increased PGE2 production which stimulates expression of the P450C17 enzyme enabling progesterone to be converted to estrogen. The estrogen formed is then responsible for the increased production of PGF2a that occurs in the maternal tissues. On-going in vivo and in vitro studies are examining this model.

Role of Drug Resistance Proteins in Pregnancy

A group of transporters in the ATP binding cassette (ABC) superfamily have recently been found in the placenta including the multidrug resistant gene product 1 (Mdr-1) also known as P-glycoprotein (P-gp) and Breast Cancer Resistance Protein (BCRP). These proteins are potentially capable of protecting the fetus from a large number of drugs and toxins. Mdr-1/P-gp is also involved in the extrusion of various steroids including certain glucocorticoids and may therefore play an important role in regulating fetal access of glucocorticoids. Studies in human placenta and the mouse are being carried out to define the expression and regulation of Mdr-1/P-gp and BCRP. We have recently shown the there is a marked decrease in the expression of placental Mdr-/P-gp in the human and mouse placenta with advancing gestation which may be regulated by the decrease in progesterone levels seen in the latter species.
